PSD-95 in CA1 Area Regulates Spatial Choice Depending on Age

چکیده
Cognitive processes that require spatial information rely on synaptic plasticity in the dorsal CA1 area (dCA1) of hippocampus. Since function hippocampus is impaired aged individuals, it remains unknown how animals make choices. Here, we used IntelliCage to study behavioral support choices female mice living a group. As proxy training-induced plasticity, analyzed morphology dendritic spines and expression scaffold protein, PSD-95. We observed choice training young adult induced correlated shrinkage downregulation PSD-95 dCA1. Moreover, long-term depletion by shRNA dCA1 limited correct reward corner, while preference was intact. In contrast, old strategies characterized an increased tendency for perseverative visits social interactions. This strategy resulted robust corner during task. decreased correlation between size spines. Furthermore, did not impair place or mice. Thus, our data indicate PSD-95-dependent choices, observe cage mates stick preferred seek reward. resistant area. Overall, demonstrates combine alternative molecular approach consume rewards complex environment. SIGNIFICANCE STATEMENT It poorly understood aging affects cognitive functions. is, however, essential understand these develop therapeutic interventions successful aging. Our (i.e., information), Second, contribution functions changes with age.

متن کاملCyclin-dependent kinase 5 regulates PSD-95 ubiquitination in neurons.
Cyclin-dependent kinase 5 (Cdk5) and its activator p35 have been implicated in drug addiction, neurodegenerative diseases such as Alzheimer's, learning and memory, and synapse maturation and plasticity. However, the molecular mechanisms by which Cdk5 regulates synaptic plasticity are still unclear. PSD-95 is a major postsynaptic scaffolding protein of glutamatergic synapses that regulates synap...
متن کاملUbiquitination Regulates PSD-95 Degradation and AMPA Receptor Surface Expression
PSD-95 is a major scaffolding protein of the postsynaptic density, tethering NMDA- and AMPA-type glutamate receptors to signaling proteins and the neuronal cytoskeleton. Here we show that PSD-95 is regulated by the ubiquitin-proteasome pathway. PSD-95 interacts with and is ubiquitinated by the E3 ligase Mdm2. In response to NMDA receptor activation, PSD-95 is ubiquitinated and rapidly removed f...

To address the unsolved question of PSD-95 function I used short hairpin RNA (shRNA) to knock-down protein expression. shRNA mediated knock-down of protein expression is one of the established methods exploiting the naturally occurring expression control mechanism known as RNA interference (RNAi). Cells express shRNAs that are complementary to the mRNA they control. The shRNAs are processed and...
متن کاملActivity-dependent regulation of synaptic strength by PSD-95 in CA1 neurons.
CaMKII and PSD-95 are the two most abundant postsynaptic proteins in the postsynaptic density (PSD). Overexpression of either can dramatically increase synaptic strength and saturate long-term potentiation (LTP). To do so, CaMKII must be activated, but the same is not true for PSD-95; expressing wild-type PSD-95 is sufficient. This raises the question of whether PSD-95's effects are simply an e...
